The emerged from Keepmoat Stadium as 3-0 victors on Wednesday, courtesy of a brace from Zimbabwe international Admiral Muskwe and a late effort from George Hirst.
City's youngsters also finished top of Northern Group A following victories over Sunderland and Grimsby Town as well as a penalty shootout success at Scunthorpe United.
Their reward for an excellent campaign so far this season will be a trip to Prenton Park to tackle Micky Mellon's Tranmere, who are sat in 20th position in Sky Bet League 1.
Leicester's Round 3 tie on Merseyside is scheduled to take place on the week commencing Monday 6 January, 2020.
